چکیده

In a recent laser light-scattering study on the “globule-to-coil” transition (melting) of a single poly(Nisopropylacrylamide) (PNIPAM) linear chain in water (the solution is in the stable one-phase region at all temperatures), we observed that at the initial melting stage the ratio of the hydrodynamic radius to the radius of gyration underwent an unexpected maximum s,1.61d which is even higher than s5y3d1y2 predicted for a rigid and uniform globule, indicating a molten globule. This finding suggests that just as with a protein molecule the melting of a homopolymer globule is not an all-or-none process and the molten state is a general phenomenon for macromolecules in solution. Globule-to-Coil Transition of a Single Homopolymer Chain in Solution

Using a nearly monodisperse high molar mass poly(N-isopropylacrylamide) (PNIPAM) sample, we successfully made the conformation change of individual PNIPAM chains from a coil to a fully collapsed stable single chain globule in an extremely dilute aqueous solution, which enabled us to study for the first time the globule-to-coil transition of a single homopolymer chain in solution. A comparison t...

Comparison of the Coil-to-Globule and the Globule-to-Coil Transitions of a Single Poly(N-isopropylacrylamide) Homopolymer Chain in Water

Using a newly prepared nearly monodisperse (Mw/Mn < 1.05) high molar mass (Mw ) 1.3 × 107 g/mol) poly(N-isopropylacrylamide) (PNIPAM) sample, we successfully, for the first time, made the conformation of individual PNIPAM chains change from a coil to a fully collapsed thermodynamically stable single chain globule and then back to a coil in an extremely dilute aqueous solution (∼6.7 × 10-7 g/mL)...

Structural Characteristics of Stable Folding Intermediates of Yeast Iso-1-Cytochrome-c

Cytochrome-c (cyt-c) is an electron transport protein, and it is present throughout the evolution. More than 280 sequences have been reported in the protein sequence database (www.uniprot.org). Though sequentially diverse, cyt-c has essentially retained its tertiary structure or fold. Thus a vast data set of varied sequences with retention of similar structure and fun...

Reentrant coil-to-globule-to-coil transition of a single linear homopolymer chain in a water/methanol mixture.

Both water and methanol are good solvents for poly( N-isopropylacrylamide) (PNIPAM) at room temperature, but strangely not a mixture of them. Using narrowly distributed linear PNIPAM chains (M(w) = 2.6x10(7) g/mol and M(w)/M(n)<1.1), we have, for the first time, observed a coil-to-globule-to-coil transition of a single PNIPAM chain when methanol was gradually added into water. This novel reentr...

Protein folding and unfolding studied at atomic resolution by fast two-dimensional NMR spectroscopy.

Atom-resolved real-time studies of kinetic processes in proteins have been hampered in the past by the lack of experimental techniques that yield sufficient temporal and atomic resolution. Here we present band-selective optimized flip-angle short transient (SOFAST) real-time 2D NMR spectroscopy, a method that allows simultaneous observation of reaction kinetics for a large number of nuclear sit...
